Description
Pierce high capacity Ni-IMAC resin contains a matrix that is a 6% crosslinked agarose with optimal properties for batch, gravity column, and research/pilot scale FPLC column purifications. The resin has high binding capacity (up to 80 mg of protein per mL of resin) and can tolerate the high EDTA and reducing agent concentrations used to inhibit metalloproteases and stabilize sensitive intracellular proteins.
Features of the high-capacity EDTA-compatible Ni-IMAC resin include:
- Chelator stable—use up to 20 mM EDTA and DTT with no loss in performance
- Compatible with purification of overexpressed, secreted proteins in cell culture media (e.g., Expi293, ExpiCHO, and ExpiSf expression systems)
- High binding capacity—>80 mg/mL binding capacity for His-tagged recombinant proteins
- Reusable—can be regenerated up to 10 times using the CIP protocol
EDTA and reducing agents such as DTT and TCEP adversely affect the performance of Ni-IMAC supports by stripping off the metal. Because of its chelator stability characteristic (EDTA tolerant), Pierce high-capacity EDTA-compatible Ni-IMAC resin is especially suited for purifying expressed His-tagged proteins that are secreted into cell culture media or for purifying intracellular His-tagged proteins that need the presence of EDTA to maintain stability and function. In both cases, large quantities of samples can be purified without the Ni metal stripping off of the support, thereby providing consistent high capacity yields.
