Description
- Recognizes human PD1
- PD1 (Programmed cell death 1, CD279) is an immunoinhibitory receptor that belongs to CD28/CTLA-4 subfamily of Ig superfamily
- Is a 288 amino acid Type I transmembrane protein composed of one Ig superfamily domain, stalk, transmembrane domain and intracellular domain containing an ITIM and ITSM
- Expressed on T cells, B cells, natural killer T cells, activated monocytes, dendritic cells
- Engagement by ligands transduces a signal that inhibits T cell proliferation, cytokine production and cytolytic function
- Critical for regulation of T cell function during tolerance, autoimmunity and infection
- Blockade of PD1 can overcome immune resistance and shown to have antitumor activity
Process:
- Antibody specific for PD1 pre-coated onto microwells
- PD1 protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for PD1 is added to detect captured PD1 protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
