Description
- Recognizes human E-Selectin
- E-Selectin (ELAM-1, CD62E) is a memeber of selectin family of adhesion molecules that include L-Selectin and P-Selectin
- E-Selectin is inducible endothelia cell surface molecule
- Can be secreted by endothelial cells and detected in soluble form
- Plays role during inflammation in recruiting leukocytes to site of injury
Process:
- Antibody specific for E-Selectin pre-coated onto microwells
- E-Selectin protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for E-Selectin is added to detect captured E-Selectin protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correctional wavelength set at 630 nm
