Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous APOA1

  • Recognizes human APOA1
  • APOA1 (apolipoprotein A1) is a plasma protein and protein component of HDL
    • Associated with reversed cholesterol transport, lipid/cholesterol binding, LCAT activation and specific receptor binding
    • Synthesizes in liver and small intestine
    • Defects of APOA1 cause low HDL level and systematic non-neuropathic amyloidosis
    • Serum concentration of APOA1 inversely related to risk of developing atherosclerosis
    • Alterations of APOA1 correlate to cancer and Alzheimer's disease

Process:

  • Antibody specific for APOA1 pre-coated onto microwells
  • APOA1 protein in samples captured by coated antibody after incubation
  • After extensive washing, another antibody specific for APOA1 is added to detect captured APOA1 protein
  • For signal development, Sterptravidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
